Small animal imaging mainly uses bioluminescence and fluorescence technologies. Bioluminescence uses the luciferase for marking, while fluorescence technology uses green/red fluorescent proteins. Fluorescent proteins require excitation light of a specific wavelength to excite fluorescence. Interestingly, animal hair and other tissues will also exhibit fluorescence, resulting in background that affects observation (By removing hair from the observation area, the background can be reduced). Most researchers still use bioluminescence technology for small animal imaging.
